A comprehensive assessment of the neurotransmitter pool in the rat brain under different condition of lead intoxication

Various toxic substances that affect the brain tissue, need a comprehensive assessment for the description of the changes in physiologically different structures: hypothalamus, middle brain, striatum and large hemispheres. In here, the lead intoxication has been put under investigation. We assessed the most important indicators characterizing changes in the pool of neuroactive compounds in different brain structures under the influence of lead acetate. We used several experimental models for analysis, e.g. models with the use of adult and junior rats, different time of pregnant female rat intoxication with further analysis of their offspring, different ways of intoxication. The levels of neuroactive amino acids as well as levels of biogenic amines and their derivatives were extracted from the brain tissues with the use of chloric or formic acids and were further analysed with the use of reverse-phase and ion-pair HPLC analysis. Data obtained in all experiments under different lead poisoning conditions were used to create a matrix of changes. The analysis made on the basis of the matrix allowed us to select one-way changes that occurred within the same brain structure in three or more cases. Such a way of data analysis enables to see general orientation of changes in the levels of specific neuroactive compounds. Thus, the matrix created as a result of our study may be used as a template for comparison of levels neuroactive compounds in different brain structures.
